Sitting down face-to-face will ensure they understand the importance of the issue and how it's affecting your work. If you work remotely and your HR team is far away, schedule a video or audio call to discuss the matter. Avoid only putting it in an email, so your tone doesn't get misunderstood.

In short — IMO, if you apply and the only contact info provided after applying is HR, then I'd say it's fair game to reach out. I would personally call rather than email though to get a feel for their *customer service*. However.... You may not get very far simply by asking for an “update” on your application.
When I have encounter a position I was interested in but wanted more details I would simply call HR and ask some questions. If they say they are not sure or I am not satisfied with their answers I ask to speak with the hiring manager.
When HR disregards your complaints, be sure to file a complaint with the Equal Employment Opportunity Commission (EEOC). The EEOC may investigate your employer and seek evidence, and may even recommend mediation so a resolution can be reached with the assistance of an objective third party.
